VEGF 配体与细胞膜上的 VEGFR 受体结合,诱导受体二聚化和激活,进而启动细胞内信号级联反应,导致血管内皮细胞增殖、存活、迁移及通透性增加(Matsumoto and Mugishima, 2006)。受体主要形成同源二聚体,但已观察到 VEGFR-1 和 VEGFR-2 之间的异二聚体。尽管 VEGFR-1 和 VEGFR-2 均在血管内皮中表达,但体内 VEGF 的血管生成活性主要通过 VEGFR-2 传递。

VEGF binds to VEGFR leading to receptor dimerization The binding of VEGF ligands to VEGFR receptors in the cell membrane induces dimerization and activation of the latter, initiating intracellular signaling cascades that result in proliferation, survival, migration and increased permeability of vascular endothelial cells (Matsumoto and Mugishima, 2006). The receptors predominantly form homodimers but heterodimers between VEGFR-1 and -2 have been observed. Although both VEGFR-1 and -2 are expressed in the vascular endothelium, the angiogenic activities of VEGFs are transduced mainly through VEGFR-2 in vivo.
