GABA 合成由两种功能不同且需要不同辅因子的谷氨酸脱羧酶（GAD65 和 GAD67）唯一介导。GAD65 与 VGAT（GABA 转运体）功能关联，且 GAD65 合成的 GABA 优先被装载入突触囊泡。GAD67 合成的 GABA 可用于除神经递质传递以外的其他功能。
