前起始复合物的形成分五个步骤进行：TFIID 识别并结合核心启动子元素，TFIIA 和 TFIIB 结合到 Pol II 启动子：TFIID 复合物，TFIIF 招募 Pol II 全酶到 Pol II 启动子：TFIID:TFIIA:TFIIB 复合物，TFIIE 结合到正在形成的前起始复合物，形成闭合的前起始复合物（Orphanides et al. 1997）。
