当神经元细胞缺乏 NTRK3(TRKC)配体 NTF3(NT-3)时,NTRK3 作为依赖性受体发挥作用,促进细胞凋亡。NTRK3 的促凋亡活性与神经系统的正常发育有关,通过决定存活的感觉神经元数量(Tauszig-Delamasure et al. 2007)。在没有配体的情况下,NTRK3 发生依赖 caspase 的切割(Tauszig-Delamasure et al. 2007),导致 NTRK3 杀手片段(KF)的释放。NTRK3 KF 与 NELFB(COBRA1)形成复合物,插入线粒体膜中,促进细胞色素 c 释放和凋亡体介导的凋亡(Ichim et al. 2013)。

NTRK3 as a dependence receptor When neuronal cells are deprived of the NTRK3 (TRKC) ligand NTF3 (NT-3), NTRK3 functions as a dependence receptor, promoting apoptosis. The pro-apoptotic activity of NTRK3 is implicated in proper nervous system development, by dictating the number of surviving sensory neurons (Tauszig-Delamasure et al. 2007). In the absence of its ligand, NTRK3 undergoes caspase-dependent cleavage (Tauszig-Delamasure et al. 2007), resulting in release of the NTRK3 killer fragment (KF). The NTRK3 KF, in complex with NELFB (COBRA1), inserts into the mitochondrial membrane, promoting cytochrome c release and apoptosome-mediated apoptosis (Ichim et al. 2013).
