De novo fatty acid biosynthesis converts acetyl-CoA into long-chain saturated fatty acids, primarily palmitate (C16:0), through a multi-step process involving the key enzymes acetyl-CoA carboxylase for malonyl-CoA production and the fatty acid synthase complex. This pathway is highly regulated by insulin, citrate, and leucine.
