Purine metabolism encompasses de novo biosynthesis of purine nucleotides (AMP, GMP) from ribose-5-phosphate via the committed step catalyzed by PRPP amidotransferase. The salvage pathways recover free purines via HPRT and APRT enzymes; defects cause Lesch-Nyhan syndrome or gout.
